The Extraction buffer is not harmful or does not contain any irritation substances.

The optimal mixture of Extraction buffers and PCR-Enzyme mix guaranties a fast and repeatable result.

Kit Components:

The Direct-PCR Kit contains:

- Extraction Buffer (EXT-buffer), 10000 µl

- Thermostable Enzyme Mix (ENZ-Mix), 500 µl

- Hot-Start PCR Mix with loading dye, 1300 µl

- Sterile Water 1500 µl

The components are enough for about 100 reactions (25µl/reaction)

Storage: -20°C for about 12 months

Transport: on blue ice

Additionally required (not included):

0,2 ml PCR Tubes, Thermoblock, Waterbath or Thermocycler, optional for swaps sample 1,5 ml Tubes,

Typical extraction protocol for e.g. Animal Tissue (for all detailes protocols please open the Direct-PCR-Kit product datasheet)

Add to a 0,2 ml PCR Tube:

- 85 µl EXT-Buffer

- 5 µl ENZ-Buffer

incubate:

10 min @ 50°C

  5 min @ 95°C

run PCR-protocol or store @ +4°C for up to 1 month.

Post PCR Analysis:

The PCR-Product contails already two dyes, blue and yellow for direct loading on the gel.

Trouble shooting:

In case of:

- lysate is too thick

- or any problem with pipetting: centrifuge and uses supernatant in PCR-
  Reaction; or dilute the lysate 1:5 to
  1:10 with TE buffer or Tris-HCl, pH 8.0.

- of non-specific or lack of PCR product dilute the lysate 1:5 to 1:10 with
  TE buffer or Tris-HCl, pH 8.0.
